Chemical investigation of the ethyl acetate extract from the twigs of Millettia leptobotrya Dunn led to the isolation of five isoflavonoids (1–5) and one phenolic derivative (6). Their structures were established through comprehensive NMR spectroscopic analyses and comparison with published literature and identified as (+)-trans-4,7,2′-trihydroxy-4′-methoxyisoflavanol (1), prunetin (2), 5-hydroxy-4′,7-dimethoxyisoflavone (3), 5,4′-dihydroxy-7,8-dimethoxyisoflavone (4), isotectorigenin, 7, 4´-dimethyl ether (5), and atraric acid (6). This study represents the first report of compounds 1, 5, and 6 from the genus Millettia, further contributing to the phytochemical characterization of this species. The cytotoxic activities of compounds 1, 4, and 5 were evaluated against KB, HepG2, A549, and MCF-7 human cancer cell lines using the MTT assay. Only compound 1 showed weak cytotoxicity against KB, HepG2, A549, and MCF-7 cancer cells, with IC50 values of 44.92, 48.70, 64.68, and 49.23 μg/mL, respectively.
