An UV-Vis absorption spectroscopy method had been developed and validated to quantify total polyphenols in the leaves of Terminalia catappa L. (TC). The TC leaf powder was extracted by ultrasonic method with 50% ethanol, followed by the reaction with Folin-Ciocalteu phenol reagent and photometric measurement at 730 nm. The developed method exhibited good linearity (R2 = 0.9957) within a range of 1-8 µg/ml and LOD and LOQ were found to be 0.033 and 0.100 µg/ml, respectively. The developed method was also found accurate, and precise according to the regulatory guidelines. The developed method was applied to measure the total polyphenol content in samples collected from various provinces in Vietnam.
