Optimization of Pressurized Solvent Extraction Using Response Surface Methodology and HPLC Analysis of Vitexin from Passiflora Foetida L.
The study aims to optimize the automated sample preparation step using accelerated solvent extraction (ASE) and response surface methodology (RSM) and integrate it with a simple, quick, precise, and accurate high performance liquid chromatography (HPLC) technique for vitexin measurement in P. foetida. The RSM results demonstrated that the optimal conditions for vitexin extraction were an extraction temperature of 150.59°C, an extraction time of 19.16 min, and a solvent volume of 140.91 mL; The actual values at the optimal conditions according to the model showed the maximum value of the objective function of vitexin yield was 11.18 ± 0.15 (mg/g). The validated HPLC method was as follows: A mixture of acetonitrile-0.1% formic acid solution with the elution gradients were: 0 - 40 min, 15% acetonitrile; 40 - 45 min, 100% acetonitrile. Other running conditions included the detection wavelength (342 nm), the flow rate (1 mL/min), the injection volume (20 µL), and the column temperature (40°C).
