Physalis angulata is an important medicinal plant that has been widely used in traditional medicine for the treatment of inflammation and various inflammatory-related disorders. Despite its medicinal relevance, comprehensive chemical profiling of the fruits of P. angulata collected in Vietnam has not yet been systematically investigated. In this study, a comprehensive chemical characterization of the methanolic extract of P. angulata was performed using high-resolution quadrupole time-of-flight mass spectrometry (HR-QTOF-MS), enabling rapid and accurate profiling of its bioactive constituents. Furthermore, the anti-inflammatory activity of the isolated compounds was evaluated by measuring lipopolysaccharide (LPS)-induced production of pro-inflammatory cytokines, including interleukin (IL)-12p40, IL-6, and tumor necrosis factor-α (TNF-α), in bone marrow-derived dendritic cells. Consistent with this, compound 1 exhibited potent anti-inflammatory activity against IL-12p40, IL-6, and TNF-α, with IC50 values of 1.17 ± 0.02, 7.52 ± 0.03, and 2.93 ± 0.05 µM, respectively, compared with the positive control adezmapimod (IC50 = 5.00 ± 0.01, 3.50 ± 0.01, and 7.20 ± 0.01 µM, respectively). Overall, this study provides the first comprehensive chemical profile of P. angulata from Vietnam and offers scientific evidence supporting its traditional use as a source of anti-inflammatory agents.
